All three of these amino acids are involved in the urea cycle and are key contributors to nitric oxide production. L-Ornithine is distinct from L-Arginine and L-Citrulline as it is not involved with nitric oxide production, but L-Ornithine buffers ammonia reducing fatigue associated with exercise. 

When your cells break down nitrogen-containing compounds such as proteins, they produce ammonia as a byproduct and can be toxic if not removed.

Ammonia Detoxification

The Urea Cycle is a system in the liver that converts nitrogen waste into urea and eliminates it from the body. The production and removal of urea is essential for the elimination of ammonia and toxic nitrogen metabolites. Ammonia can be extremely toxic to cells and excessive levels can cause serious symptoms and disease.  While urea cycle disorders are often caused by genetic diseases, deficiencies of arginine or ornithine triggered by conditions such as illness or stress can cause increases in blood ammonia levels. Even in healthy athletes, the build-up of ammonia can lead to extreme fatigue and dementia because it reduces glycogen formation and inhibits the energy cycle.

Nitric Oxide Production

Nitric oxide is essential to relaxing blood vessels. Nitric oxide helps the blood vessels maintain their flexibility so that the blood flow is unrestricted. This, in turn, helps maintain healthy blood pressure and improved blood flow to organ systems. Nitric oxide helps prevent blood clots because it prevents blood aggregation or platelets from becoming sticky. When platelets are not sticky, they can move in a single file through the capillaries. Additionally, nitric oxide works as an antioxidant that reduces the possibility of immune cells adhering to artery walls. This helps keep down inflammation, which most health professionals agree is a major cause of the plaque formation that contributes to atherosclerosis.


L-Ornithine is called a nonessential amino acid because your body can manufacture it. As part of the urea cycle, your liver cells produce ornithine from the amino acid arginine, which you consume in many different foods. Dietary sources of ornithine include meat, fish, eggs and dairy products. Some studies suggest that taking ornithine prior to consuming alcohol may reduce hangovers due to alcohol increasing ammonia levels. This may be particular to those with decreased liver function or those with alcohol sensitivity.

L-Arginine is the only substrate in the biosynthesis of nitric oxide (NO), which plays critical roles in important physiological processes in the human body including neurotransmission (nerve communication between cells), vasorelaxation (dilation of blood vessels), protective cytotoxicity (removal or bad cells) and immunity. L-Arginine is essential for the muscle cells to produce creatine, a compound needed for muscle contraction. It also facilitates the release of growth hormone, insulin, glucagon, and prolactin, important hormones for metabolic drive. In the cardiovascular system, due to the vasorelaxation, arginine supplementation can cause a reduction in systolic and diastolic pressures in patients with high blood pressure, provides a positive effect on brain circulation and can enhance blood flow to males with erectile dysfunction. Arginine has also been found to support a reduction in total cholesterol and decreased low density lipoproteins (LDL).

L-Citrulline is important in vasodilation, the widening of blood vessels, resulting from relaxation of smooth muscle cells within the vessel walls. L-Citrulline's benefits stem primarily from its ability to increase levels of Arginine. In the body, citrulline is converted to the amino acid arginine, thus raising plasma and tissue levels of arginine and enhancing nitric oxide production.
